The first written mention of the church dates back to 1582, in connection with the transfer of the burgher Gapka Tabkova in the ownership of the Trinity Church of his estate.
During the Turkish rule in the city, the temple was converted into a mosque. At the beginning of the XVIII century. with the withdrawal of the Turks from Kamianets and the establishment of Polish rule, the church was transferred to the Uniates. In 1722, Bishop Athanasius Sheptytsky handed over the church to the monks of the Order of St. Basil, who founded a monastery there. The official date of foundation is May 22.1722. The Order of Basilians operated at the church until 1795. During the Second Partition of Poland, the lands of Kamianets-Podilskyi were annexed by Russia. At the same time, the church was reconsecrated to the Orthodox Church.

In 1935, as part of the Soviet government's struggle against religion, the church was dismantled.

On March 3.1995, the restoration of the church began. Excavations and research of the foundations of the temple were carried out. On September 23.1996, Cardinal Lubomir Husar founded the monastery.
In five years, the Basilians completely completed the reconstruction of the building and decoration of the temple and the surrounding area.
